SS31, also known as Elamipretide, is a novel, cell-penetrating aromatic-cationic peptide at the forefront of mitochondrial research. Unlike most peptides that interact with surface receptors, SS31 is designed to target the primary source of cellular energy: the mitochondrion.
Its mechanism of action centers on its high affinity for cardiolipin, an exclusive phospholipid of the inner mitochondrial membrane. In conditions of high oxidative stress, cardiolipin is susceptible to damage, which destabilizes mitochondrial structure and impairs the electron transport chain (ETC), leading to reduced ATP (energy) production and a cascade of further oxidative damage.
SS31 selectively binds to cardiolipin, acting like a molecular chaperone to protect it from oxidative degradation. By stabilizing the cardiolipin-ETC interface, SS31 is reported to help restore the efficiency of oxidative phosphorylation, enhance ATP synthesis, and decrease the production of reactive oxygen species (ROS).
