SS-31, also known as Bendavia or MTP-131, is a synthetic, cell-permeable aromatic-cationic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2. Its unique structure allows it to freely cross cell membranes and selectively accumulate within mitochondria, independent of the mitochondrial membrane potential.
The primary mechanism of SS-31 involves its interaction with cardiolipin, a phospholipid exclusively found on the inner mitochondrial membrane. By binding to cardiolipin, SS-31 acts as a powerful antioxidant, scavenging harmful reactive oxygen species (ROS) like hydrogen peroxide and hydroxyl radicals directly at their source.
This targeted action protects mitochondrial integrity, enhances ATP (energy) production, and inhibits the opening of the mitochondrial permeability transition pore (mPTP). By preventing mPTP opening, SS-31 effectively blocks a key pathway to apoptosis (programmed cell death) and cellular damage.
